§2What this calculator assumes

  • That the labelled mass is delivered. It generally is not: peptide content, residual water and counter-ion each reduce the delivered peptide mass below the label, and none is visible from a purity figure. The size of that discrepancy can be worked through in the purity-against-content calculator.
  • That the diluent volume added is the volume measured. Reconstitution into a lyophilisate cake displaces a small volume, which this calculator does not model.
  • That the syringe is a U-100 insulin syringe, graduated in units of 0.01 mL.
  • That no peptide is lost to container or syringe surfaces. At concentrations below roughly 0.2 mg per millilitre and in the absence of a surfactant, adsorption removes a measurable fraction. The Institute cannot quantify that term without compound-specific data and does not attempt to.
